Description
This study aimed to determine the phenolic compound profile of the methanol extract of Bornmuellera cappadocica (Willd.) Cullen & T.R.Dudley (Brassicaceae) and to comparatively evaluate the enzyme inhibitory activities of the extract and the silver nanoparticles (BCAgNPs) green-synthesized using this extract. The phenolic compound profile of the methanol extract was determined using liquid chromatography-tandem mass spectrometry (LC–MS/MS). The enzyme inhibitory activities of the extract and BCAgNPs were assessed based on half-maximal inhibitory concentration (IC₅₀) values determined against carbonic anhydrase-II (CA-II), acetylcholinesterase (AChE), butyrylcholinesterase (BChE), and α-glucosidase enzymes. LC–MS/MS analysis revealed the quantitative presence of five phenolic compounds in the methanol extract. p-Coumaric acid was identified as the predominant phenolic compound, with a concentration of 231.911 µg/g of extract, followed by chlorogenic acid (184.613 µg/g) and resveratrol (76.381 µg/g). The total content of the identified phenolic compounds was calculated to be 559.847 µg/g of extract. The high levels of p-coumaric and chlorogenic acids, in particular, demonstrated that the extract possesses a noteworthy composition regarding hydroxycinnamic acid derivatives. In enzyme inhibition assays, the IC₅₀ values of the methanol extract against CA-II, AChE, BChE, and α-glucosidase enzymes were determined to be 1.126 mg/mL, 0.00304 mg/mL, 0.00754 mg/mL, and 0.0275 mg/mL, respectively. For BCAgNPs, the IC₅₀ values for the same enzymes were found to be 0.102 mg/mL, 0.125 mg/mL, 0.145 mg/mL, and 0.00013 ± 0.0000163 mg/mL, respectively. These results indicate that BCAgNPs exhibited higher inhibitory activity than the methanol extract against CA-II and α-glucosidase enzymes, but lower inhibitory activity against AChE and BChE enzymes. In conclusion, it was demonstrated that the B. cappadocica methanol extract contains various phenolic compounds primarily p-coumaric acid and chlorogenic acid and that both the extract and the green-synthesized silver nanoparticles exhibit varying inhibitory activities depending on the enzyme type.
